Medical Practical Report – Acid Phosphatase Kinetics
Paper details:
Introduction (approx. 500 words – 25%): This should contain some background on phosphatases, and the acid phosphatase family of enzymes. The phosphatase used in our practical was isolated from wheat germ. Feel free to include any information that you feel is relevant, but remember that the information in the intro should ideally give the reader a suitable preface to – and a contextualisation of – your results and discussion (though it does not have to be limited to such information). Links to material on acid phosphatase can be found at the end of this document.
Methods (approx. 500 words – 25%): The methods section should contain an overview of the procedures you used in the lab, and a brief explanation of why each experiment was done, what controls were used and why? Can you think of any additional positive or negative controls that the experiments would have benefited from?
Results (approx. 500 words – 25%): This should include tables and bar charts or line graphs of all results obtained, with suitable legends. All results should be displayed and described but not discussed. These should include the Lineweaver-Burke plot, your results for Km and Vmax, as well as the workings of the calculations used to obtain these values.
Discussion (approx. 500 words – 25%): Discuss the results obtained for all variables. Wherever you can, describe whether the data obtained point to any optimum conditions for enzyme activity, and try to relate this to enzyme biology where possible. Are there molecular biochemical explanations for the observed effects of pH alteration on active site function and thus enzyme activity? Is there any precedent for such effects already published in the scientific literature and if so, how do your results compare with these findings?
